- 20 ~ 80mm : gram-positive
- 7 ~ 8 mm : gram-negative

24hours cultured
E.Coli in medium

Crystal violet

Gram iodine

95% ethyl-alcohol

Safranin solution
(25% safranin in 50% ethyl-alcohol)

Wash the slide glass with water quickly and blot dry gently.

Smear is now ready to be read with microscope. (Use oil immersion lens.)
